First, before anything else, please try updating your graphics drivers, you want whatever driver is the most up-to-date version available.
Then, to address the mouse situation, try disabling high dpi resizing:
1. Right click on the game in Steam
2. Click on "Properties"
3. Go to the "Local Files" tab and select "Browse local files" This should bring up the location of the game on your computer.
4. Right click on "FTLGame" (the application)
5. Select "Properties"
6. Click on the "Compatibility" tab
7. Tick the box next to "Disable display scaling on high DPI settings"
8. Click "Ok" to save the change.
If you still can't click anything to change the settings, you can try editing settings.ini manually.
To start, try editing the "#Manual resolution choices - overrides anything in the options" to set the values there to match your screen size. E.g. -
